GHK-Cu
GHK-Cu is a naturally occurring copper-binding peptide widely used as a cosmetic skincare ingredient, with small human studies on topical use and no meaningful evidence for injected use.
GHK-Cu is a very small molecule: three amino acids - glycine, histidine and lysine - bound to a copper ion. The tripeptide GHK occurs naturally in human blood plasma, and it was first identified in the 1970s by researchers studying differences between plasma from younger and older donors. GHK has a strong affinity for copper, and the copper-bound complex is the form usually referred to as GHK-Cu. Copper itself is an essential trace element involved in a number of enzyme systems, including enzymes that cross-link collagen and elastin in the skin.
GHK-Cu is unusual among the compounds covered on this site because it has a long and entirely legitimate commercial history as a cosmetic ingredient. It appears in a great many topical skincare products sold openly in the UK and elsewhere, where it is regulated as a cosmetic rather than as a medicine. That gives it a different profile from research-chemical peptides - but it also creates confusion, because the evidence supporting a cosmetic topical ingredient does not transfer to injected use, and the two are often discussed as if they were the same thing.
How it is thought to work
The proposed mechanism has two main strands. The first is copper delivery. Copper is required by lysyl oxidase, an enzyme involved in cross-linking collagen and elastin fibres, and by superoxide dismutase, an antioxidant enzyme. The hypothesis is that GHK acts as a carrier that can deliver copper into tissue in a controlled way, since free copper ions are reactive and not well tolerated. The peptide-bound form is proposed to be a gentler vehicle.
The second strand concerns gene expression. Laboratory work, largely in cultured human skin cells, has reported that exposure to GHK-Cu is associated with changes in the expression of a large number of genes, including genes involved in the production of extracellular matrix proteins such as collagen. This work is real and has been published in peer-reviewed journals, but it should be read for what it is: observations in cells in a dish. A change in gene expression in cultured fibroblasts is a starting point for investigation, not a demonstration that anything visible happens to a person's skin.
What the research actually shows
GHK-Cu sits higher on the evidence ladder than the purely preclinical peptides, but not as high as popular marketing implies. The foundation is a substantial body of cell culture and animal work on wound models and collagen synthesis. Above that there are small human studies - the important qualifier being small. Several trials of topical GHK-Cu creams have been published, typically enrolling a few dozen participants, often over periods of weeks to a few months, and reporting outcomes such as measured skin roughness, density or appearance. Some of these were controlled and some were funded by parties with a commercial interest in the ingredient, which is a standard limitation in cosmetic research and worth knowing about.
What this amounts to is early-human evidence for topical cosmetic use: enough to justify the ingredient's presence in skincare, not enough to make strong claims. There are no large, long-duration, independent randomised controlled trials establishing the size or durability of any effect, and the studies that exist use varied formulations and varied outcome measures, which makes them hard to compare. When a product page states a specific percentage improvement, that number almost always traces back to a single small study with its own methodology, not to a settled scientific consensus.
Injected GHK-Cu is a completely different matter, and this is the point where the online conversation departs from the evidence entirely. The human studies that exist looked at topical application to skin. There is no comparable body of human research on injecting GHK-Cu. Claims about systemic effects - on organs, on overall ageing, on tissue repair beyond the skin - are extrapolated from cell culture and animal data, not from human trials of injected use. There is an additional consideration specific to this compound: it delivers copper, and copper is a trace element with a narrow range between sufficiency and excess. Systemic copper loading is not a trivial matter, and the human data needed to characterise that risk from injected GHK-Cu has not been generated.
Where the evidence stops
- Human trials are small, short and heterogeneous in formulation and outcome measures.
- Several studies were commercially sponsored, and independent replication at scale is limited.
- Effect sizes reported for cosmetic outcomes are modest and not consistently measured between studies.
- Topical evidence says nothing about injected use - the two routes are not interchangeable.
- No meaningful human evidence exists for any systemic or anti-ageing effect beyond the skin.
- The consequences of systemic copper delivery from injected use are uncharacterised in humans.
- Cell culture gene expression findings have not been shown to translate into measurable human outcomes.
Legal and regulatory status
In the United Kingdom, GHK-Cu is legal and common as a cosmetic ingredient in topical skincare products. Cosmetics are regulated under UK cosmetics legislation, which requires a safety assessment and appropriate labelling but does not involve the efficacy testing required of a medicine. That is why cosmetic products may describe appearance-related effects but may not make medicinal claims - a cosmetic is regulated on the basis that it works on the surface, not that it treats a condition.
GHK-Cu is not an approved medicine in the UK. There is no MHRA marketing authorisation for it as a treatment for anything, and injectable GHK-Cu is not an approved product. Material sold in vial form for injection is generally supplied as a research chemical labelled not for human consumption, and carries none of the manufacturing, sterility or purity assurances that apply to either a licensed medicine or, in its own more limited way, a regulated cosmetic. The legality of the cosmetic ingredient should not be read as regulatory endorsement of the injected form: they are different products under different rules.
